Does it matter that the two tees supplying am additional rad(a small one for an airing cupboard) on a one pipe system, are only 6 inches apart?

Yes it does.
The pipe should run under the rad with the tees looking straight up into the valves, preferably TBOE. Flow at the top.
If your thinking of using TRVs they must be suitable for a one pipe system.
